Does the enhanced serial tower board (TWR-SER2) make high-speed (HS) USB possible with the Kinetis K22 tower board (TWR-K22F120M)?

The K22 has full speed OTG and not HS USB so it can't be used with the HS tranceiver on the SER2 board.
You will need a K26 for HS USB.
