I want to use Azure IoT middleware for my application with Kinetis K64 controller but having following questions.

How much flash memory is required to run azure IoT application ?

what is the size of Azure IoT stack with umqtt after build it ?

Hope you are doing well. 

I recommend to read the following post from the Azure website on the recommendation for minimum space needed.

I'd recommend to test the examples provided in the SDK. The amount of memory it covers will be listed in the .map file.
