We have made a custom board from IMX8MP.
In the board there is no USB OTG available.
USB hosts are available though.
Is there any way to do DDR calibration using MScale DDR Tool, without USB OTG?

No, it is not possible as specified in the DDR stress tool user guide:
It is a program running on the PC side which downloads a test image to the i.MX series processor’s internal RAM through a USB connection.

I can see MScale DDR Tool is only available in .exe format. Is there any variant for Linux OS?

This tool support DDR stress test for i.MX8M family devices.
