I've been asked by a third party to authenticate a sector of a Mifare Plus 2K EV1 card with a 16-byte B key (instead of the typical 6-byte).

Is this possible?  If I understand correctly, sector trailers are still the same format and length as Mifare Classic. The only part I'm unsure of is the SL1 functionality and AES-128 (which would be 16-bytes).

The rest of the card is in transport configuration and can be read using the same authentication as Mifare Classic which is why I assume it could be SL1 (and not SL3) but I'm a bit fuzzy on if AES-128 is just used at the wireless communication level or if it's a replacement for sector authentication?
